These are averages across a trial population. Some people lost considerably more, others much less, and results depend on staying on treatment.

Orforglipron side effects

Most side effects hit the stomach and gut, and they are strongest in the first weeks and after each dose increase. Reported ones include nausea, constipation, diarrhoea, vomiting, indigestion, stomach pain, bloating, belching, heartburn, gas, headache and tiredness. Some people notice hair thinning. Smaller meals and drinking enough fluid help most of it settle.

Foundayo carries a boxed warning for thyroid tumours, including a rare thyroid cancer seen in animal studies. The label also flags pancreatitis, gallbladder problems, severe stomach reactions, kidney injury caused by dehydration from heavy vomiting or diarrhoea, serious allergic reactions, and worsening of diabetic eye disease in people with type 2 diabetes. Low blood sugar becomes more likely if you also take insulin or a sulfonylurea, so those doses may need adjusting.

Get medical help quickly

Severe or lasting stomach pain, especially pain spreading to your back, can signal pancreatitis. A lump or swelling in the neck, a hoarse voice or trouble swallowing also need same-day attention. So does dehydration from repeated vomiting, or confusion and sweating that point to a sugar crash.

Who should not take orforglipron

It is ruled out if you or a close family member has had medullary thyroid cancer, if you have the condition MEN2, or if you have had a serious allergic reaction to it before. It is not approved for anyone under 18.

You have had pancreatitis before, you have severe gastroparesis where the stomach empties too slowly, or you have kidney problems. Pregnancy matters here too.
